After the historic launches earlier this year of the first two HCV-specific protease inhibitors, expectations were high at the American Society for the Study of Liver Disease’s (AASLD) 2011 Liver Meeting where numerous developers presented promising data for HCV-specific Phase II and III antivirals. Among the agents striving to be included in the first interferon-free regimens, Pharmasset’s nucleoside polymerase inhibitor PSI-7977, Bristol-Myers Squibb’s NS5A inhibitor daclatasvir, Boehringer Ingelheim’s protease inhibitor BI-201335 and Novartis/Debiopharm’s alisporivir grabbed the limelight at the conference. Encouraging Phase III trial results for these therapies strengthen the growing recognition that highly effective, safe and convenient interferon-free regimens may be available in the near future. The company that successfully brings the first interferon-free combination to market will be well-positioned to capitalize on the significant population of diagnosed patients who are unable or unwilling to undergo interferon-based treatment.

At the Liver Meeting, trial data were also presented from studies evaluating HCV antivirals in specific patient subpopulations. These include interferon-based studies of Vertex’s Incivek and Merck/Roche’s Victrelis in HIV/HCV co-infected individuals and Medivir/Johnson & Johnson’s second-generation protease inhibitor simeprevir in cirrhotic genotype 1-infected nonresponders.

About the Speaker

Seamus Levine-Wilkinson, Ph.D., is an analyst in Decision Resources’ infectious disease group. He earned a doctorate in cellular, molecular and developmental biology and biophysics at Johns Hopkins University. In his dissertation, he focused on the host cell biology of Salmonella-induced tubules and incorporated live-cell imaging studies of invasive Salmonella in epithelial cells.
